چکیده

Soybean cotyledonary cells harvested every 5-10 days at 15 to 60 days after flowering (OAF), were investigated by means of light and transmission electron microscopy. In the early developing stages (15-20 OAF) most of the cells were occupied by large, centrally located vacuoles while the cytoplasm was restricted to a thin layer against the cell wall and contained numerous ribosomes, mitochondria, plastids, small amounts of endoplasmic reticulum (ER) and minute lipid bodies. At about 25 OAF, spherical organelles which contained protein, lipid and sugar (PLS bodies), appeared and then increased in number and in size. Vacuoles had protein deposits lining the inner surfaces of the tonoplasts. At 30-35 OAF, the PLS bodies lost their characteristic ciccular shape because of invaginations containing cytoplasmic materials and small vacuoles . Such transformed PLS bodies, fragmented vacuoles and protein bodies became d ifficult to distinguish from eac h other. At this stage, the cytoplasm contained abundant rough ER with cisternae, dictyosomes , lipid bodies and plastids . At 35-40 OAF, protein bodies which vari ed in size and shape were observed in ce lls a long with lipid bodies and rough ER. At about 40 OAF the plastids reached maximum size and number and then most disappeared . In the final stage ( 55-60 OAF) , protein bodies became homogeneous in electron density with completion of protein accumulation. Studies on lipid synthesis and degradation in developing soybean cotyledons.

The metabolic activity of individual lipid classes found in developing soybean cotyledons (Glycine max.) is estimated by determining the degradation rate of the compound under given conditions. Pulse-labeling and dual substrate labeling are used to evaluate this parameter. These studies indicate first order decay kinetics for phosphatidic acid, phosphatidylinositol, phosphatidylcholine, phospha...

Involvement of phospholipids in triglyceride biosynthesis by developing soybean cotyledons.

The incorporation of phospholipids specifically labeled with glycerol-2(3)H and acyl-(14)C by whole cell tissues of developing soybean cotyledons (Glycine max L.) reveals that phosphatidylinositol, phosphatidylcholine, phosphatidylethanolamine, N-acylphosphatidylethanolamine, and phosphatidic acid can be metabolized to diglyceride. The diglyceride formed may be recylced into phospholipid or acy...

Expression of a gene for uricase II (nodulin-35) in cotyledons of soybean plants.

A cDNA clone (URcot-35) was isolated from a soybean cotyledonary cDNA library using a cDNA clone (URnod-35) for nodule uricase II as a probe. URcot-35 was a 1,170-bp cDNA with an open reading frame that encoded a protein of putative 309 amino acids with a molecular mass of 35,137 Da. The nucleotide sequence of URcot-35 was identical to that of URnod-35. Expression of the URcot-35 gene in cotyle...

Arginase is inoperative in developing soybean embryos.

Arginase (EC 3.5.3.1) transcript level and activity were measured in soybean (Glycine max L.) embryos from the reserve deposition stage to postgermination. Using a cDNA probe for a small soybean arginase gene family, no transcript was detected in developing embryos. However, arginase transcripts increased sharply on germination, reaching a maximum at 3 to 5 d after germination. There was low bu...

Energetics of sucrose transport into protoplasts from developing soybean cotyledons.

The accumulation of tetraphenylphosphonium (TPP(+)), 5,5'-dimethyl-oxazolidine-2,4-dione (DMO), and a micro pH electrode were used to measure membrane potential, intracellular and extracellular pH, respectively, upon the addition of exogenous sucrose to soybean cotyledon protoplasts. Addition of sucrose caused a specific and transient (a) depolarization of the membrane potential (measured by TP...
